HIV感染后CD4^+ T细胞表面NK相关受体表达的变化 被引量:4

Expressions of natural killer cell receptors on CD4^+T cells of HIV-infected individuals

摘要 目的研究人类免疫缺陷病毒(human immunodeficiency virus,HIV)感染后CD4+T细胞表面NK相关受体表达的变化。方法选取25例未经高效抗逆转录病毒治疗的HIV感染者、11例AIDS患者、15例进行高效抗逆转录病毒治疗(highly activeantiretroviral therapy,HAART)者和13例HIV抗体阴性健康对照,用流式细胞仪检测研究对象外周血CD4+T细胞表面NKG2D、NKG2A和KIR3DL1的表达。结果 AIDS患者CD4+T细胞表面NKG2D表达的百分比显著高于其他各组,且NKG2D表达的百分比与CD4+T细胞的绝对数量呈负相关(R=-0.352,P<0.05),与HIV病毒载量呈正相关(R=0.426,P<0.05)。AIDS患者CD4+T表面NKG2A表达的百分比显著高于其他各组,NKG2A+NKG2D-表达的百分比显著高于其他各组,且NKG2A表达的百分比与CD4+T细胞的绝对数量呈负相关(R=-0.432,P<0.01)。结论 HIV感染机体后,CD4+T细胞NK相关受体表达变化与疾病进展相关。 In order to study the alteration of natural killer cell-associated receptors on CD4+T cells of HIV infected individuals,peripheral blood was collected from 25 HIV infected individuals without treatment(HIV group),11 patients in AIDS stage(AIDS group),15 HIV infected patients with highly active antiretroviral therapy(HAART group) and 13 normal controls with negative HIV antibody(NC group).The expressions of NKG2A,NKG2D,and KIR3DL1 on CD4+ T cells surfaces were analyzed with flow cytometer.We found that the expression of NKG2D on CD4+T cells in AIDS group was significantly higher than those in other groups,while the expression of NKG2D+ on CD4+T cells was negatively correlated with absolute the count of CD4+T cells(R=-0.352,P 0.05) but positively correlated with viral load(R=0.426,P 0.05).Furthermore,the expressions of NKG2A and NKG2A+ NKG2D-on CD4+T cells in AIDS group were significantly higher than those in other groups,and the expression of NKG2A+ on CD4+T cells was negatively correlated with absolute the count of CD4+T cells(R=-0.432,P 0.01).Our results indicate that the expressions of natural killer cell receptors on the surfaces of CD4+T cells during HIV infection are related with disease progression.
