摘要
目的:通过检测趋化因子CCR2,CXCL9在早、中、晚期口腔黏膜下纤维性变(OSF)组织中基因及蛋白表达的改变,进一步探讨OSF的发病机制。方法:收集早、中、晚期OSF组织,RT-PCR检测趋化因子CCR2,CXCL9基因mRNA的表达,Western blot检测组织中趋化因子CCR2,CXCL9蛋白的表达。结果:趋化因子CCR2,CXCL9基因mR-NA及蛋白在早、中、晚期OSF组织中与对照组相比均呈现表达下调,在早、中、晚期病变组织中表达依次递减。在早期OSF病变组织中,CCR2,CXCL9基因mRNA较健康对照组分别上调406%和2051%,蛋白表达分别升高385%和1506%;在中期OSF病变组织中,CCR2,CXCL9基因mRNA较健康对照组分别上调369%和1901%,蛋白表达分别升高310%和1419%;在晚期OSF病变组织中,CCR2,CXCL9基因mRNA较健康对照组分别上调357%和1760%,蛋白表达分别升高309%和1401%。结论:早、中、晚OSF各期组织中趋化因子CCR2,CXCL9基因mRNA和蛋白水平表达均异常升高。此类因子的异常表达可能与OSF的发病机制相关。
Objective: To further explore the pathogenesis of oral submucous fibrosis (OSF) by testing the expressions changes of chemokine CCR2,CXCL9 in early, metaphase and advanced stage of OSF. Method:The expressions of chemokine CCR2, CXCL9 were detected by RT-PCR and Western blot techniques. Result:To compare to the control group, the mRNA of chemokine CCR2, CXCL9 increased by 406 % and 2051%,and the protein expression increased by 385 % and 1506 % in early stage of OSF;the mRNA of chemokine CCR2,CXCL9 increased by 369 % and 1901%,and the pro- tein expression increased by 310 % and 1419 % in metaphase stage:the mRNA of chemokine CCR2,CXCL9 increased by 357 % and 1760 %,and the protein expression increased by 309 % and 1401% In advanced stage. Conclusion:The gene and protein expressions of chemokine CCR2, CXCL9 were highly upregulated in different stages of OSF.The expressions changes of them maybe in relation of the pathogenesis of OSF.
