FANCF在宫颈癌的表达缺陷与其临床特征关系的研究 被引量:1

摘要 目的:研究FANCF基因在宫颈癌的表达缺陷与高危型HPV感染及宫颈癌临床特征的关系。方法:逆转录聚合酶链技术(RT-PCR)及Western Blot分析检测60例宫颈癌组织标本及60例正常宫颈组织中FANCF mRNA及蛋白的表达,并检测高危型HPV感染情况。结果:49例(81.7%)宫颈癌组织中FANCF mRNA和蛋白表达缺陷(下降或缺失)。宫颈癌组织与正常宫颈组织相比,FANCF mRNA、蛋白相对表达值均下降(0.56±0.12vs1.48±0.09和0.16±0.03vs0.98±0.04),差异均有统计学意义(P<0.05)。FANCF表达缺陷与宫颈癌患者FIGO临床分期、WHO分级、肿瘤大小及脉管转移、高危型HPV感染有关。结论:宫颈癌患者FANCF表达缺陷。FANCF表达缺陷对宫颈癌发生、进展有一定的作用。FANCF表达缺陷与高危型HPV感染可能在宫颈癌的发生中起协同作用。 Objective:To evaluate the expression of FANCF gene in cervical cancer and to analyze its relationship with the clinical characteristics of cervical cancer. Methods:The expressions of FANCF mRNA and protein were detected by reverse transcription polymerase chain reaction (RT-PCR) and Western Blot respectively in sixty cervical cancer specimens and sixty, normal cervical tissues. Results:The expressions of FANCF m RNA in cervical cancer tissues decreased ( 0.56 ±0.12) significantly or absolutely defaulted (49/ 60) compared to normal cervical tissues ( 1.48 ±0.09 ), and the expressions of FANCF protein also decreased in cervical cancer (0.16 ±0.03) when compared normal cervical tissues ( 0.98 ±0.04 ), There were statistically significant differences ( P 〈0.05 ). Decreased expressions of FANCF were also related to the clinical stage, the tumor size, the metastasis, the infection of the high risk human papilloma virus (HPV). Conclusions: Decreasing expression of FANCF gene plays an important role in the development of cervical cancer which may also has obvious synergistic effect with HPV infection in the development of cervical cancer.
