摘要 目的研究维生素E琥珀酸酯(VES)对人食管癌Eca-109细胞系增殖、凋亡及相关蛋白表达的影响。方法实验组为不同浓度VES,对照组为同等体积RPMI-1640培养基,分别作用24、48和72 h后,MTT测细胞增殖;电镜观察细胞凋亡的形态学;流式细胞术(FCM)测凋亡率及survivin、caspase-3蛋白表达。结果各实验组抑制率随VES处理浓度的增加、作用时间的延长而逐渐升高(P<0.01)。实验组细胞呈现不同程度凋亡改变,且随VES浓度增加,凋亡率逐渐升高(P<0.05)。各实验组survivin蛋白表达减少(P<0.05),caspase-3蛋白表达增加(P<0.05)。Survivin与caspase-3表达呈负相关(r=-0.95,P<0.05)。结论 VES可抑制Eca-109细胞增殖、诱导凋亡,可能与下调survivin蛋白和上调caspase-3蛋白表达有关。 Objective To study the effects of Vitamin E Succinate (VES) on proliferati sions of associated protein of human esophageal carcinoma cell line Eca-109. Methods on, apoptosis and expres- Experimental groups were VES of different contrations and control group was culture medium in same volume. After Eca-109 cells were ex- posed for 24,48 and 72 h, MTT was used to measure the proliferation of cells. Morphological changes of Eca-109 cells were observed by transmission electron microscopy. Apoptotic rate and the expression of survivin, caspase-3 protein of Eca-109 among different concentration groups was examined by flow cytometry. Results The inhibitory rate increased gradually with the increasing concentration of vitamin E succinate and prolonging of treatment time (P 〈 0. 01 ). Apoptosis appeared in different degree in cells. With the increasing concentration of VES, the apoptot- ic rate of every treatment group was sion of survivin in Eca-109 reduced significantl graduately y higher than that of control group (P 〈 0. 05 ). The protein expres- ( P 〈0. 05 ) , but caspase-3 increased gradualy ( P 〈0. 05 ). two proteins had inverse correlation( r = - 0. 95, P 〈 0. 05 ). Conclusions Vitamin E succinate can inhibite These prolif-eration and induce apoptosis of Eca-109 cell line. The effect may be ralated to the up-regulation of caspase-3 protein expression and down-regulation of the survivin expression.
