摘要
对北京地区24个杏鲍菇栽培品种的遗传多样性进行RAPD分析,选取了40条RAPD随机引物对供试的24个杏鲍菇菌株的基因组DNA进行PCR扩增,最终成功筛选出9条RAPD引物,共扩增出142条稳定、清晰的DNA条带;聚类分析结果表明,在相似系数为0.850的水平时,可将24个供试菌株分为5大类;主坐标分析结果与聚类分析基本一致;RAPD分子标记技术成功的表明了北京地区杏鲍菇菌株的遗传多样性,为杏鲍菇的遗传育种研究奠定基础。
The aim of this study was to obtain the genetic diversity of 24 kinds of Pleurotus eryngii in Beijing region. The genetic relationships among 24 P. eryngii strains were analyzed by RAPD with 40 random primers. Finally, 142 specific and stable amplification patterns were obtained by using 9 primers. A dendrogram constructed by using the amplification data separated the test strains into five groups at a coefficient of 0.850. Relationship among the 24 genotypes of P. eryngii visualized by Principal Coordinate Analysis (PCO) was consitent to clustering analysis. Data obtained in this study showed that RAPD could reveal the genetic diversity of P. eryngii strains in beijing region. These results laid the founda- tion for the genetic breeding of P. eryngii.
