摘要
目的:探讨动脉粥样硬化闭塞症(ASO)兔模型TNF-α mRNA、NF-κB mRNA表达的变化,部分阐明ASO炎症反应机制。方法:建立动脉粥样硬化闭塞症动物模型,观察主动脉内皮组织TNF-α mRNA,NF-κB mRNA表达情况,采用SPSS 11.0统计软件进行单因素方差检验。结果:通过以目的基因的相对转录量为参数进行统计分析可知,模型组兔子动脉内皮组织TNF-α mRNA、NF-κB mRNA的表达与对照组比较均显著升高,有统计学差异(P<0.01)。结论:ASO形成过程中,动脉内皮组织TNF-α mRNA、NF-κB mRNA的表达显著升高,提示抑制TNF-α mRNA表达,阻断NF-κB的激活,或抑制NF-κB mRNA,对于拮抗ASO的发生发展都有着极其重要的临床意义。
Objective:To observe the level changes of TNF-α mRNA and NF-κB mRNA on the established process of the atherosclerotic occlusive disease(ASO)models.And to clarify the mechanism of inflammation on the ASO disease.Methods:ASO animal model was established to observe the changes of TNF-α mRNA and NF-κB mRNA on the endarterium and make single factor analysis of variance test analysis test with SPSS 11.0 software.Results:Compared with the control group,the expression levels of TNF-α mRNA and NF-κB mRNA were markedly increased in model group.there was significant difference between the two groups(P0.01).Conclusion:The high levels of TNF-α mRNA and NF-κB mRNA on the established process of the atherosclerotic occlusive disease(ASO)show restraining the expression of TNF-α mRNA,interdicting the activation of NF-κB or restraining the expression of NF-κB mRNA may control the atheromatous plaque formation on the atherosclerotic occlusive disease.
