摘要
同源盒基因 (homeobox genes)在植物 ,动物 ,真菌的广泛存在说明这种结构在真核生物进化过程中高度保守 ,并暗示其具有重要功能 .本文将玉米 kn- 1同源盒插入表达载体 p GEMΔ Xba ,并将形成的重组克隆p GEMΔ MK转化 BL 2 1(DE3)而获得高效表达 ;表达产物的分子量为 2 9× 10 3,并主要以可溶性蛋白形式存在 .Kn-1,Quox\|1,OSIH- 1间的交叉免疫反应证实三者在蛋白质水平具同源性 .而 Kn- 1同源结构域 (Homeodomain)
Homeobox genes widespread in plants, animals and fungi and are very conserved in eukaryotic evolution. These show homeobox genes play important roles in biological evolution.In this paper, the homeodomain encoded by kn\|1 homeobox of maize is overproduced in a recombinant expression vector pGEMΔMK in E. Coli, as a form of soluable protein with a molecular weight of 29×10 3. Crossed\|immunoreaction studies among Kn\|1,Quox\|1 and OSIH\|1 indicate all three are homologous at the protein level. The Kn\|1 homeodomain is able to identify the homeotic target genes.
