摘要
目的观察丹曲林对骨骼肌再灌注损伤的保护作用及可能机制。方法SD大鼠随机分为3组:假手术组、再灌注组及丹曲林组,再灌注后,检测血清TNF-α、IL-10、MDA及组织MPO,并观察组织学变化。结果再灌注组的MDA、MPO和TNF-α增高,丹曲林组与之相比明显降低;丹曲林组的肌损伤和白细胞聚集程度与再灌注组相比明显改善;假手术组和再灌注组IL-10水平较低,给予丹曲林则显著上调IL-10水平。结论丹曲林能够减轻再灌注骨骼肌的炎症损伤,其机制可能是下调TNF-α表达并诱发IL-10的增多。
Objective To investigate the protective effect and probable mechanism of dantrolene on the skeletal muscle ischemia reperfusion injury.Methods SD rats were randomly divided into sham group,reperfusion group,dantrolene group.Serum TNF-α,IL-10,MDA,tissue MPO as well as histological change were measured after reperfusion.Results Levels of MDA,MPO,and TNF-α increased in the reperfusion group,whereas the relevant expressions in the dantrolene group decreased significantly.Histological examination demonstrated significant improvements between the same both groups.IL-10 reflected the protection observed above with a significant up-regulation of expression after dantrolene.Conclusion The ryanodine receptor antagonist dantrolene offered significant protection from the inflammatory injury of the skeletal muscle ischemia reperfusion.The probable mechanism was the down-regulation of the expression of TNF-α and the increment of IL-10.
