摘要
目的观察康脑液对脑缺血再灌注大鼠胶质纤维酸性蛋白(GFAP)和神经元核抗原(NeuN)表达的影响。方法将大鼠随机分为5组:假手术组、模型组、高、中、低剂量康脑液组;用线栓法制备大鼠局灶性脑缺血再灌注模型;再灌注后不同时间,用免疫组化方法检测GFAP、NeuN的表达。结果脑缺血再灌注后第3,7,14,21 d,康脑液组可减少GFAP的表达,与模型组相比差异有统计学意义(P<0.05,P<0.01),至缺血后21d,康脑液组GFAP表达已接近正常;康脑液组的NeuN表达增高,与模型组比较差异有统计学意义(P<0.05,P<0.01)。结论康脑液可抑制缺血侧大脑皮质GFAP的表达,诱导神经干细胞向神经元分化,促进神经的再生及修复。
Objective To observe the effect of Kangnaoye on the expression of glial fibrillary acidic protein(GFAP)and neuronal nuclear antigen(NeuN) in rats with cerebral ischemia and reperfusion.Methods These rats were randomly divided into 5 groups: sham-operated group,model group,high-dose kangnaoye group,middle-dose Kangnaoye group and low-dose kangnaoye group.The model of focal cerebral ischemia and reperfusion was induced in rats with the intraluminal suture method.The expression of GFAP and NeuN were determined by immunohistochemistry method at different time after cerebral ischemia.Results The groups treated with Kangnaoye had a lower expression of GFAP than the model group(P0.05 or P0.01) at day 3,day 7,day 14 and day 21 after cerebral ischemia.Until the day21 after cerebral ischemia and reperfusion,the expression of GFAP of Kangnaoye group approximated the level of normal.Compared with model group,the expression of the NeuN of groups treated with Kangnaoye were significantly higher than the model group(P0.05 or P0.01).Conclusion Kangnaoye can inhibit the over expression of GFAP,promote neural stem cell differentiation into neurons,and advance nerve regeneration and repair.
