摘要
目的观察脾虚证大鼠血清甲状腺激素水平的改变及针刺配合西洋参汤对其改变的影响。方法将40只wistar大鼠随机分为正常对照组、模型组、西洋参组、针药并用组,每组10只。模型组、西洋参组、针药并用组用偏食法建立脾虚证动物模型。造模成功后,模型组用生理盐水1 mL/100 g体重灌胃;西洋参组给予西洋参汤1 mL/100 g灌胃;针药并用组采用针刺双侧足三里、三阴交,并给予西洋参汤1 mL/100 g灌胃。以上治疗均为每日1次,治疗10 d。第11天处死4组动物,取样做测定血清三碘甲状腺原氨酸(T3)、甲状腺素(T4)、反三碘甲状腺原氨酸(rT3)、促甲状腺素(TSH)含量,并测量各组动物造模前后体重,胸腺、脾脏质量。结果模型组大鼠造模后体重、胸腺质量/体重、脾脏质量/体重比及血清T3、T4含量显著下降,与正常对照组比较,差异均具有统计学意义(P<0.05,P<0.01)。西洋参组与针药并用组大鼠胸腺质量/体重及T3、T4含量较模型组有显著提高(均P<0.01);针药并用组大鼠体重和脾脏质量/体重与模型组比较,差异均具有统计学意义(P<0.05)。结论针药并用能改善脾虚证大鼠的脾虚症状,其作用机理与调节甲状腺激素水平有关。
Objective To observe changes in serum thyroid hormone levels and the effect of acupuncture plus American ginseng decoction on the changes in rats with spleen deficiency syndrome. Methods Forty Wister rats were randomly allocated to normal, model, American ginseng and combined acupuncture and medicine groups, 10 rats each. An animal model of spleen deficiency syndrome was made by dietary bias in the model, American ginseng and combined acupuncture and medicine groups. After successful model making, the model group received an oral gavage of normal saline 1 ml/100 g; the American ginseng group, an oral gavage of American ginseng decoction 1 ml/100 g; the combined acupuncture and medicine group, acupuncture at bilateral points Zusanli and Sanyinjiao plus an oral gavage of American ginseng decoction 1 ml/100 g. The above treatments were given once daily for 10 days. The four groups of rats were sacrificed at 11 days. Samples were taken for the measurement of serum triiodothyronine (T3), thyroxine (T4), reverse triiodothyronine (rT3) and thyroid stimulating hormone (TSH) contents. Body, spleen and thymus weights were also measured in every group of rats before after model making. Results Body weight, thymus weight/body weight and spleen weight/body weight, and serum T3 and T4 contents decreased markedly in the model group of rats after model making. There were statistically significant differences compared with the normal group (P〈0.05,P〈0.01). Thymus weight/body weight, and serum T3 and T4 contents increased significantly in the American ginseng and combined acupuncture and medicine groups of rats compared with the model group (all P〈0.01). There were statistically significant differences in body weight and spleen weight/body weight between the combined acupuncture and medicine group and the model group (P〈0.05). Conclusions The combined use of acupuncture and medicine can relieve the symptoms of spleen deficiency in rats with spleen deficiency syndrome. The mechanism of its action is related to regulating thyroid hormone levels.
