摘要
目的研究心力衰竭(heart failure,HF)大鼠外周血生长分化因子-15(growth differentiation factor-15,GDF-15)含量变化及心肌组织GDF-15mRNA表达与心力衰竭发生发展关系。方法结扎大鼠左前降支制备心力衰竭模型,采用酶联免疫方法与实时荧光定量方法检测外周血中GDF-15及心肌组织GDF-15mRNA表达;彩色多普勒检测实验大鼠心功能。结果随着病程进展,HF大鼠外周血液GDF-15含量1452.03±76.88pg/ml高于正常大鼠外周血GDF-15含量26.53±34.06pg/ml,HF大鼠心肌组织GDF-15mRNA亦随之增高,而假手术组则表达降低,差异有统计学意义;心功能(Left ventricμlar ejection frac-tion)EF值随着外周血及心肌组织中GDF-15表达水平增高而降低。结论外周血中GDF-15含量变化及心肌组织mRNA表达调控与HF发生发展密切相关,可以作为预测心力衰竭进展新的生物标志物。
Objective To research the expressing level of GDF - 15 in peripheral blood and its mRNA in the myocardial tissue in rats in the HF development. To explore the corelation between the GDF - 15 expressing level and the development of HF in rat. Methods The establishment of rat HF model by ligaturing left descending branch of rat coronary artery :detecting GDF -15 expression level in pe- ripheral blood by enzyme linked immunosorbent assay with real -time fluorescence quantitative method,observe GDF - 15 mRNA in myo- cardial tissue by PCR,and detect the rat cardiac function by Color Doppler. Results With the development of this HF, the level of pe- ripheral blood GDF- 15 of HF group rats was 1452.03±76.88pg/ml,while it was 26.53±34.06pg/ml in normal control group rats . GDF - 15 mRNA expression in myocardial tissue shows heigher and lower level in HF group rats and normal control group rats , respective- ly,and the difference had statistical significance. EF was reduced accompanied with the increasing expression of peripheral blood GDF - 15 and tissue GDF - 15 mRNA in HF group rats. Conclusion The changes of peripheral blood GDF - 15 and the myocardial tissue GDF - 15 mRNA expression level has a close relationship with the occurrence and development of HF. The results shows that GDF - 15 is a new potential biological marker to predict heart failure progress.
