摘要
目的采用Real-time PCR方法来检测miRNA-7在小鼠12种不同组织器官中的表达情况并探讨其意义。方法首先分别提取小鼠12种组织器官总RNA,用miRNA-7特异性引物逆转录为cDNA,采用Real-timePCR探针法检测miRNA-7在12种组织器官中的表达水平。结果成功应用Real-time PCR方法检测miRNA-7在小鼠12种组织器官中的表达水平,结果显示,miRNA-7在肺组织中的表达水平最高,而在淋巴结中的表达水平最低。此外,在小肠、脑、胸腺、脾脏中也存在miRNA-7的高表达。结论 miRNA-7在小鼠的肺、小肠、脑、胸腺、脾脏中呈较高水平的表达,提示该分子可能在上述器官的发育、分化、功能维持及相关疾病发生中具有重要作用,为后续深入研究miRNA-7的生物学功能提供了前期试验依据。
Objective To establish Real-time PCR method for detecting the expression of miRNA-7 in different murine organs and explore its significance.Methods Total RNAs were extracted from 12 different murine organs,and reverse transcripted into cDNA.The expression of miRNA-7 in 12 different murine organs were detected by Real-time PCR assay.Results miRNA-7 expression could be detected in all of 12 murine organs by Real-time PCR assay.However,the level of miRNA-7 expression in lung was highest compared with that in other organs.In contrast,the level of miRNA-7 expression in lymph nodes was lowest.moreover the high level of miRNA-7 was measured in small intestime brain thymus and spleen.Conclusion miRNA-7 is enriched in lung,small intestine,brain,thymus and spleen,suggesting that miRNA-7 might play an important role in the development,differentiation,functional sustainment and the occurrence of related diseases in these organs.
