人骨髓间充质干细胞在体内向造血细胞分化的研究

摘要 目的探讨人骨髓间充质细胞(hMSC)在体内向造血细胞分化的潜能和造血重建能力。方法首先建立hMSC的分离培养扩增体系,以hMSC为供体细胞,亚致死剂量射线照射后的裸鼠为受体,通过病理、免疫组化、流式细胞术等方法观察hMSC在体内向造血细胞分化能力。结果将体外分离扩增的hMSC移植给亚致死剂量照射后的裸鼠,移植后第6周外周血白细胞明显恢复并接近正常水平。移植后8周裸鼠骨髓细胞增生活跃,可见各个阶段的细胞。通过流式细胞术、免疫组化的方法在骨髓中可以检测到CD34和CD45阳性的细胞,且能够形成造血祖细胞集落(CFU-GM)。结论 hMSC能在亚致死剂量射线照射的裸鼠骨髓中植入并分化表型为人CD34+的造血干/祖细胞。 Objective To explore the efficiency of the hematopoietic reconstuction and the proficiency of the hematopoietic differentiation of human mesenchymal stem cells(hMSC) in vivo.Methods The hMSC derived from adult bone were isolated,culture-expanded efficiently in vitro.hMSC as donor cells,sublethally irradiated nude mice as receptor,to observe the hMSC ability of hematopoietic differentiation by pathology,immunohistochemistry and flow cytometry.Results hMSC were transplanted to sublethally irradiated nude mice.After 6 weeks peripheral leukocytes was significantly recovered to near normal levels.After 8 weeks bone marrow proliferated well,and all stages of the cells were visible.CD34 and CD45 positive cells in the bone marrow can be detected by flow cytometry and immunohistochemistry,and the hematopoietic progenitor cells can form hematopoietic stem/progenitor cells colony(CFU-GM).Conclusion The hMSC can transplant in the bone marrow and differentiate into CD34+ hematopoietic stem/progenitor cells in sublethally irradiated nude mice.
