摘要
目的:建立去卵巢大鼠骨质疏松症的动物模型,观察强骨胶囊对其血清中骨保护素(OPG)、核因子-κβ受体活化因子配基(receptor activator of nuclear factor-kappaβligand,RANKL)、雌激素水平、胫骨干骨密度及胫骨生物力学的影响。方法:将3月龄48只SD雌性大鼠按照1:3的比例随机分为空白组、手术模型组。手术模型组制备成为去势大鼠模型,3个月后再将手术模型组大鼠进一步随机分为3组:强骨胶囊组、尼尔雌醇组、模型对照组,分别予以强骨胶囊、尼尔雌醇、蒸馏水(每组12只)进行灌胃3个月,3个月后处死空白组和手术模型组大鼠,取大鼠双侧胫骨及血液标本,检测治疗前后大鼠的胫骨骨密度、血清OPG、RANKL值,雌激素水平、胫骨生物力的变化。结果:造模后3个月手术模型组与空白组相比较骨密度显著下降(P<0.05);经过3个月的治疗后,其中强骨胶囊组、尼尔雌醇组与模型对照组大鼠血清OPG、RANKL含量及大鼠各部位骨密度相比有统计学意义(P<0.05)。结论:强骨胶囊对细胞因子轴OPG/RANKL/RANK系统有调节作用,能提高去卵巢大鼠胫骨骨密度及血清OPG含量,降低去卵巢大鼠血清RANKL浓度,同时改善胫骨生物力学性能。
Objective:To establish ovarieetomized osteoporosis animal model, and to observe the influence of Qianggu capsules on the serum bone protection element (OPG), receptor activator of nuclear factor-kappa β ligand (RANKL), estrogen level, bone mineral density (BMD) and biomechanies of tibia. Methods:Forty-eight female Sprague-Daw (SD) rats of three months were randomly divided into blank group and operation model group according to the proportion of 1:3. The rats of operation model group were ovariectomized. The operation model rats were further randomly divided into Qianggu capsule group, nylestriol group, and model control group (n=12) three months later. They were intragastric administration with Qianggu capsule, nylestriol, and distilled water three months three months later. The rats of blank group and operation model group were sacrificed. The changes of BMD and biomechanics of tibia, serum level of OPG, RANKL, and estrogen were detected before and after treatment. Results:The BMD of model group was significantly lower than blank group three months after established model (P〈0.05). The serum level of OPG and RANKL, and the BMD in Qianggu capsule group and nylestriol group were statistically difference compared with model group (P〈0.05). Conclusion.. Qianggu capsule can regulate the cytokine axis of OPG/RANKL/RANK system, improve BMD and biomechanics of tibia and serum level of OPG, and reduce serum level of RANKL in ovariectomized rats.
