摘要
目的:通过研究骨肉瘤细胞不同细胞株的细胞迁移能力及其血管扩张刺激磷蛋白(VASP)的表达水平,探讨骨肉瘤细胞内VASP的差异性表达与骨肉瘤细胞迁移能力的关系。方法:细胞"划痕损伤"实验(2D)和Tran-swell实验(3D)分别检测骨肉瘤细胞不同细胞株MG-63和SAOS-2的迁移能力的差异;RT-PCR分别检测这两种细胞VASP mRNA的表达水平。采用SPSS13.0软件进行统计学处理。结果:骨肉瘤细胞MG-63细胞株的的迁移能力明显高于SAOS-2细胞株,有统计学差异(P<0.05)。骨肉瘤细胞MG-63细胞株和SAOS-2细胞株内均有VASP的表达,MG-63细胞株的VASP表达水平明显高于SAOS-2细胞株。结论:VASP的高表达水平可能在骨肉瘤细胞的迁移过程中发挥着重要作用,与骨肉瘤的转移和侵袭能力密切相关。
Objective: To investigate the role of the cytoskeleton associated protein vasodilator-stimulated phosphoprotein(VASP) in the migration of human osteosarcoma cells.Methods: We analyzed the cell migration ability of human osteosarcoma cell lines MG-63 and SAOS-2 by wound-healing assay(2D) and transwell assays(3D migration).We then detected the mRNA expression levels of VASP of SAOS-2 as well as MG-63 by RT-PCR.Results: The study revealed that the high metastatic potential osteosarcoma cell line MG-63 showed a higher migration ability and higher VASP expression level as compared with the low metastatic potential of SAOS-2.Conclusion: The higher expression level of VASP contributes to a higher cell migration capacity of human osteosarcoma cells.
