硼替佐米对前列腺癌细胞抑制作用及其机制

摘要 目的:探讨硼替佐米对前列腺癌细胞DU145的抑制作用。方法:采用MTT法检测硼替佐米对DU145细胞的生长抑制作用,流式细胞技术测定细胞周期和凋亡率,Western blot检测Bik和Caspase-3的表达水平的变化。结果:硼替佐米对DU145细胞的生长抑制作用有时间和浓度的依赖性。1.6μmol/L的硼替佐米作用DU145细胞后,可见明显细胞核凝聚、皱缩和碎裂;细胞的周期处于G0/G1和G2/M期的细胞比例上升,S期的细胞比例下降(P<0.05);细胞凋亡率明显增加,显著高于对照组(P<0.05),Bik和Caspase-3表达水平上调。结论:硼替佐米可明显抑制前列腺癌细胞DU145的生长并诱导其凋亡,凋亡作用机制可能与其上调Bik和Caspase-3表达有关。 Objective: To investigate the inhibitory effect of bortezomib on the prostate cancer cells DU145.Methods: MTT method was used to detect the proliferation of DU145 cells after exposed to various concentration of bortezomib.The apoptosis and cell cycle of cells were tested by flow cytometry(FCM).The expression of Bik and Caspase-3 was measured by Western blot.Results: Bortezomib inhibited the proliferation of DU145 cells in a time-and dose-dependent manner.After treatment with 1.6 μmol/L bortezomib,DU145 cells showed obvious nuclear condensation,chromatin condensation and fragmentation.Analysis of cell cycle showed that the percentage of cells in G0/G1 and G2/M phases were increased and decreased in S phase cells(all P0.05).The apoptosis rate of DU145 cells induced by bortezomib was increased and higher than that of control group,with up-regulated expression of Bik and Caspase-3.Conclusion: Bortezomib could obviously enhance the inhibitory effects and apoptosis of DU145 cells.The possible mechanism may be the upregulation of Bik and Caspase-3 expression.
