枸杞多糖对小鼠脑缺血再灌注损伤后氧化应激的影响 被引量:8

摘要 目的观察枸杞多糖对小鼠脑缺血再灌注引起的氧化应激的抑制作用。方法以枸杞多糖10、20、40 mg/kg灌胃给药,1次/d,连续7 d,于末次给药1 h后采用大脑中动脉阻塞再灌注法(MCAO/R)制作小鼠局灶性脑缺血再灌注损伤模型。采用酶学方法测定缺血侧脑组织中谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、乳酸脱氢酶(LDH)的活性及三磷酸腺苷(ATP)、丙二醛(MDA)含量。结果与模型组比较,枸杞多糖可提高脑缺血再灌注损伤后小鼠脑组织中GSH-Px、SOD、CAT、LDH酶活性及ATP含量,降低MDA含量(P<0.05)。结论枸杞多糖对脑缺血再灌注损伤后脑组织的保护作用可能与抗氧化应激、提高能量代谢有关。 Objective To investigate the possible mechanism of protective effect of lycium barbarum polysaccharide (LBP) on berebral ischemic reperfusion injury in mouse. Methods LBP was administered intragastrically for seven days by 10, 20 and 40 mg/kg. Model was established through the cerebral artery occlusion/reperfusion (MCAO/R) at lh after the last giving. Spectrophotometric as- say was used to determine the activities of glutathione peroxidase (GSH -Px), superoxide dismutase (SOD), catalase (CAT) and lac- tate dehydrogenase ( LDH), and the content of adenosine triphosphate (ATP) and malondialdehyde (MDA). Results LBP significant- ly increased activities of SOD, GSH - Px, CAT and LDH and decreased the contents of MDA in ischemic reperfusion cerebral tissue ( P 〈 0.05, P 〈 0.01 ). Conclusion The protective effect of LBP on the cerebral ischemia and reperfusion injury may be connected with the effect of anti - oxidative stress and improvement of energy metabolism.
