补骨脂乙素诱导HepG2细胞凋亡及对Bcl-2家族表达的影响 被引量:8

摘要 目的:观察补骨脂乙素对人肝癌细胞HepG2凋亡及其相关蛋白的影响。方法:用不同浓度补骨脂乙素(3、4、5、6和7mg/ml)处理HepG2细胞24h后,MTT法检测药物对细胞增殖的影响,并用流式细胞仪检测细胞凋亡情况,Western印迹法检测凋亡相关蛋白的表达。结果:补骨脂乙素4mg/ml即能抑制HepG2细胞的增殖并诱导其凋亡,7mg/ml时凋亡率可达94.57%,在浓度4~7 mg/ml之间有良好的量效关系。补骨脂乙素浓度为7mg/ml时促凋亡蛋白Bax、Bid表达明显增加、抗凋亡蛋白Bcl-2表达减少。结论:补骨脂乙素4mg/ml能诱导HepG2细胞凋亡,可能与其影响Bcl-2家族蛋白的表达有关。 Object:To investigate effect of corylifolinin to apoptosis in HepG2 cells and expression of the related protein.Method:Except for the control group,the other groups were given corylifolinin(3,4,5,6 and 7mg/ml).After 24 hours'cultivation,the drug effect on proliferation of HepG2 was detected by MTT,the condition of cell apoptosis was tested by fiow cytometry.And the expressions of apoptosis related proteins were detemined by Western blot.Result:When the dose of corylifolinin is 4mg/ml,it could inhibit the proliferation of HepG2 cells and induce apoptosis of which.The dose of 7 mg/ml of corylifolinin could induce 94.57% of HepG2 cella apoptosis.The rate of apoptosis showed a significant dose-effect relationship between 3 mg/ml to 7 mg/ml.When the dose of corylifolinin is 7mg/ml,it could increase the expression of pro-apoptotic protein Bax、Bid,but decrese the expression of anti-apoptotic protein Bcl-2.Conclusion:When the dose of corylifolinin is 4mg/ml,it could cause HepG2 cells apoptosis through influencing the protein expression of Bcl-2familiy.
