Anti-inflammatory effects of FK1 suppository in vaginitis

摘要 目的:观察FK1栓对实验性阴道炎的抗炎作用。方法:①采用苯酚胶浆致兔的阴道炎模型,观察并记录阴道的充血、水肿程度,组织病理变化;②采用大鼠蛋清和二甲苯致炎,测量大鼠足肿胀及小鼠耳肿胀的程度;③通过小鼠皮肤损伤实验,观察损伤组织愈合程度;④采用磷酸组胺致豚鼠搔痒实验计算致痒阈;⑤通过体外蛋白沉淀实验,观察蛋白沉淀程度。结果:①给药大鼠阴道充血、水肿程度明显改善,分泌物消失,红肿、充血程度明显减轻,FK1栓高、中、低剂量组(33.6mg/kg、16.8mg/kg、8.4mg/kg)家兔血白细胞数量有不同程度的下降,与模型组相比有显著性差异(p<0.05、p<0.01、p<0.001),且呈剂量依赖性。病理结果:高剂量组炎症明显好转,接近正常组,中、低剂量组炎细胞数量较模型组明显减少;②FK1栓高剂量(140mg/kg)组大鼠足肿胀程度有明显减轻并且FK1栓高剂量(200 mg/kg)对二甲苯引起的小鼠耳肿胀有抑制作用,与模型组比较差异显著(p<0.05);③FK1栓各组(100mg/kg、50mg/kg、25 mg/kg)伤口愈合面积均较模型组明显(p<0.05、p<0.01、p<0.001);④FK1栓各组(110.0mg/kg、55.0mg/kg、27.5 mg/kg)的致痒阈均高于模型组,具有显著性差异(p<0.05、p<0.01、p<0.001);⑤FK1栓高、中剂量组(240、120mg/ml)各管蛋白完全沉淀,FK1栓低剂量组(60mg/ml)各管蛋白1/2沉淀。结论:FK1栓具有抗炎消肿、收敛止痒、促进伤口愈合作用。 Objective:To observe anti-inflammatory effects of FK1 suppository in vaginitis.Methods:Rabbit vaginitis model was induced by phenol slurry,the alteration of organization hyperemia,edema,and histopathology were observed and recorded.Egg white and xylene induced proinflammatory model in rats and mice was conducted respectively,the variation of swelling on rat paw and mouse ear was calculated.Skin trauma experiment was carried out to observe the changes of healing process.Histamine phosphate itching experiments in guinea pigs was executed to examine the itch threshold.A protein precipitation experiment in vitro was performed to observe the level of protein precipitation.Results:The congestion and edema in vagina was significantly improved after FK1 treatment compared to model group.Meanwhile,the secretions was also disappeared in FK1 treatment groups.The white blood cell number was remarkably decreased in FK1 suppository treatment groups(33.6mg/kg,16.8mg/kg,8.4mg/kg) compared to the model group(p0.05,p0.01,p0.001 respectively) in a dose-dependent manner.Histopathological results showed Infiltration of inflammatory cells was inflammation of was significantly improved in the FK1 high dose(33.6mg/kg) group,which presented closely as control group,infiltration of inflammatory cells of in FK1 16.8mg/kg and 8.4mg/kg treatment groups were also significantly reduced compared to the model group.Foot swelling degree of FK1(140mg/kg) treatment group in rats was significantly decreased compared to model group.Furthermore,the swelling in ear of mice was also remarkably decreased in 200 mg/kg FK1 treatment compared to ear swelling model group(p0.05).In the Skin trauma experiment,FK1(100mg/kg,50mg/kg,25 mg/kg) treatment groups significantly improve the wound healing in a dose dependent manner compared to model group(p0.05,p0.01,p0.001 respectively).Itch threshold in Histamine phosphate itching experiments was significantly increased in FK1(110.0mg/kg 55.0mg/kg,27.5 mg/kg) treatment groups compared to model group(p0.05,p0.01,p0.001,respectively).Moreover,results from in vitro protein precipitation experiment showed that there was no sediments presented in FK1 240,120 mg/ml treatment group,and there half protein sediment in 60mg/ml FK1treatment.Conclusion:the FK1 bolt has significant anti-inflammatory effect,which presented as decreasing swelling,depressing itch feeling,promoting wound convergence and healing.
