p53凋亡刺激蛋白——一个新的肿瘤抑制家族的研究进展 被引量:1

摘要 p53凋亡刺激蛋白家族(Apoptosis-stimulating of p53 protein,ASPP)是近年来新发现的蛋白家族,包括3个成员:ASPP1、ASPP2和iASPP。ASPP蛋白在C端具有保守的锚蛋白重复序列、SH3结构域和脯氨酸富集区。ASPP是首个被发现的能调节p53家族所有成员转录活性的蛋白家族。ASPP1和ASPP2可以特异性增强p53诱导促凋亡基因表达的能力,而iASPP会抑制p53的转录活性,对多种癌细胞的研究发现,AS-PP的异常表达与癌细胞抗肿瘤药物耐受性有关。此外,ASPP在细胞极性、器官形成和IGF信号通路中也起着重要作用。该文结合最新研究进展,对ASPP家族蛋白的结构和功能作一综述。
