摘要
根据已报道的水仙潜隐病毒(Narcissus latent virus,NLV)外壳蛋白(Coat protein,CP)基因序列,设计一对特异性引物,以提取的水仙样品总RNA为模板进行RT-PCR,并进行特异性和灵敏度测定。结果表明,此法能够从携带NLV的水仙样品中成功扩增出大小约829 bp的特异性目的片段,具有良好的特异性和较高的灵敏度,可有效地应用于水仙样品中NLV的检测。
RT-PCR was established to detect Narcissus latent virus(NLV)using a pair of specific primers which were designed and synthesized based on the published nucleotide sequence of NLV. The cDNA was synthesized from the total RNA prepared from narcissus sample and then PCR was carried out. The expected 829-bp product was amplified from the NLV-infected narcissus sample. The results showed that RT-PCR method had good specificity and high sensitivity. It could be concluded that the method could be effectively applied to the detection of NLV in narcissus samples.
