摘要
目的应用微小RNA(miRNA)芯片技术筛选心脏成纤维细胞(cardiacfibroblasts,CFs)及心脏肌成纤维细胞(cardiacmyofibroblasts,CMFs)中差异表达的miRNAs,为寻找与心脏纤维化有关的miRNA提供理论基础。方法体外分离培养小鼠心脏成纤维细胞,常氧培养传代至第2代,然后在37℃、3%O2条件下培养48h得到心脏肌成纤维细胞。免疫荧光染色鉴定心脏成纤维细胞和心脏肌成纤维细胞,miRNA芯片技术分别检测miRNA在心脏成纤维细胞及心脏肌成纤维细胞中的表达。结果芯片实验数据分析软件V.4.0对芯片数据进行聚类分析,筛选获得80个在心脏成纤维细胞及心脏肌成纤维细胞中差异表达的miRNAs。相对于心脏成纤维细胞,心脏肌成纤维细胞中有55个miRNAs表达显著上调,25个miRNAs表达显著下调。结论心脏成纤维细胞和心脏肌成纤维细胞中存在差异表达的miRNA分子,差异表达可能与心脏纤维化的发生发展有关。
Objective To screen and identify the microRNA(miRNA) differential expression profiles in cardiac fi- broblasts and cardiac myofibroblasts by the miRNA microarray technique. Methods Cardiac fibroblasts isolated from neonatal mice were cultured in vitro at 37 ℃ under normal oxygen, cells were used at the second passage for the following experiments ,then cultured at 37℃ in 3% 02 for 48 h to obtain cardiac myofibroblasts. The cardiac fibroblasts and cardiac myofibroblasts were identified by immunofluorescent staining, the miRNAs were measured by miRNA chip technology in cardiac fibroblasts and cardiac myofibroblasts. Results By analysis of software V. 4.0 based on microarrays screening, 80 miRNAs was significantly altered between cardiac fibroblasts and cardiac myofibroblasts. In the cardiac myofibro- blasts, compared with cardiac fibroblasts, 55 miRNAs were significantly up - regulated and 25 miRNAs were significantly down- regulated. Conclusions miRNAs are differentially expressed between cardiac fibroblasts and cardiac myofibro- blasts, which may be related to the pathogenesis and progression of cardiac fibrosis.
