瘦素诱导肝星状细胞增殖及银杏叶提取物调控作用的研究

摘要 目的:观察银杏叶提取物(GbE)对外源性瘦素(Leptin)诱导肝星状细胞(HSC)增殖功能的影响。方法:分别以不同浓度瘦素(0、10、50、100、200、400μg/L)在不同时相(6、12、18、24h)对复苏后处于对数生长期的HSC-T6细胞进行刺激,同时采用银杏叶提取物(20、40、80、160mg/L)对100μg/L瘦素刺激的HSC-T6细胞作用,设常规培养组为对照,均用四甲基偶氮唑盐(MTT)比色法检测细胞的增殖情况。结果:不同剂量瘦素在不同时间点对HSC-T6细胞增殖呈现剂量、时间依赖性(P<0.05)。银杏叶提取物能明显抑制瘦素刺激的HSC-T6的增殖(P<0.05)。结论:瘦素可明显刺激HSC的增殖,GbE对瘦素刺激的HSC的活化途径的抑制,可能是其抗肝纤维化的重要机制。 Objective: To observe the effects of Ginkgo biloba extract (GbE) on exogenous leptin induced hepatic stellate cell (HSC) proliferation. Methods: HSC-T6 cells were treated with different concentrations of leptin (0,10, 50,100,200,400txg/L) at different phases (6,12,18,24 h). GbE (20,40,80,160mg/L) was cultured with 1001~g/L leptin stimulated HSC-T6. The control group was cultured with conventional method. MTY assay was used to detect cell proliferation in both groups. Results: The stimulation of leptin on the proliferation of HSC-T6 was in a dose-and time-dependent manner (P〈0.05). GbE significantly inhibited leptin-stimulated proliferation of HSC-T6 (P〈0.05). Conclusions: Leptin can stimulate the proliferation of HSC. GbE can inhibit the stimulation. This may be the underlying mechanism of GbE anti-hepatic fibrosis.
