丙酮酸乙酯对氯气染毒大鼠肺组织一氧化氮及一氧化氮合酶抑制作用 被引量:2

Effect of EthylPyruvate on Nitric Oxide and Nitricoxide Synthase in Lung Induced by Chlorine Gas

摘要 目的探讨丙酮酸乙酯(EP)对氯气染毒大鼠肺组织中一氧化氮(NO)含量及一氧化氮合酶(NOS)活性的影响。方法 30只SD大鼠随机分为正常对照组,染毒组和EP治疗组,每组各10只动物。正常对照组以空气为对照。染毒组给予2 536 mg/m的氯气动态染毒,染毒时间为20 min。染毒后立即给予生理盐水对照或EP(40 mg/kg)治疗。于染毒后6 h麻醉后采集样本进行肺组织病理检测,并测定组织匀浆中NO含量以及总一氧化氮合酶(TNOS)和诱导型一氧化氮合酶(iNOS)活性。结果氯气中毒后病理检查结果显示肺组织呈急性肺损伤(ALI)表现,肺组织中NO含量以及总TNOS和iNOS活性显著增加。EP治疗后显著改善了氯气中毒导致的ALI,降低了肺组织中NO含量以及总TNOS和iNOS活性。结论 EP治疗可以有效缓解氯气中毒导致的ALI,这可能与其抑制NOS活性,降低NO含量有关。 Objective To study the effect of ethyl pyruvate (EP) on nitric oxide (NO) and nitricoxide synthase (NOS) in the lung from rats exposed to chlorine gas. Methods Adult male SD rats (200 +20g, about 8 weeksof age) were exposed to chlorine gas of 2 536 mg/m or normal air for 20 min in a whole - body dynamic exposure chamber. EP (40mg/kg) or vehicle (saline) was given by intraperitoneal injections immediately after chlorine gas exposure. Rats were killed at 6 h after chlorine gas exposure, and histologic analysis, the concentration of NO and the activity of NOS were detected. Results EP attenuated pulmonary edema, hemorrhage andinflammatory cells infiltration, decreased the concentration of NO and the activity of NOS in the lung induced by chlorine gas. Conclusion EP treatment can auenuate ALI caused by the concentration of NO pate this process. chlorine gas. in lung by EP The inhibition of possibly participate this process.
