摘要
为了深入了解piggyBac转座子家族的生物学特性及分子进化特点,采用简并PCR、反向PCR、RT-PCR和RACE等方法在农业害虫亚洲玉米螟中克隆到2个piggyBac类似因子,分别命名为OfPLE1和OfPLE2。利用生物信息学软件分析Of-PLE基因编码区序列结构以及与其他物种的系统发育关系,结果表明,OfPLE1全长1 866 bp,编码607个氨基酸;OfPLE2全长1 724 bp,编码434个氨基酸。两者同源性高达70%,与粉纹夜蛾piggyBac同源性分别为32%和26%,亲缘关系较远。
The piggyBac transposon which was identified from the cabbage looper Trichoplusia ni,is a highly useful transposon in insect genetic manipulation.To further understand the biological and evolutionary characteristics of piggyBac-like elements(PLEs)in insects,two PLE sequences,named as OfPLE1 and OfPLE2,were cloned from the Asian corn borer based on degenerate PCR,inverse PCR,RT-PCR and RACE.The sequence structure of OfPLE gene coding region and the phylogenetic relationship with other species were analyzed by application of the bioinformatics software.The results showed that OfPLE1 was 1 866 bp long encoding 607amino acid residues and OfPLE2 was 1 724 bp long encoding 434 amino acid residues.Furthermore,they shared 70% homology with each other,and had 32% identity and 26% identity with Trichoplusia ni piggyBac transposon.
