摘要
目的观察心肌营养素-1(CT-1)在急性肝衰竭大鼠中的动态变化及意义探讨。方法大鼠腹腔注射D-氨基半乳糖(D-Gal)建立急性肝衰竭模型,在造模后6 h、12 h、24 h、48 h、72 h、120 h、168 h分别留取肝组织,用RT-PCR法检测肝脏CT-1mRNA的表达,并用免疫组织化学方法检测环氧化酶-2(COX-2)蛋白的表达。结果 CT-1在正常大鼠中表达很少,造模后12 h开始表达增多(P<0.05),于48 h时达高峰值,随后逐渐下降;COX-2在正常组未见蛋白表达,造模后6 h表达量逐渐增多,至48~72 h时达高峰。结论 CT-1参与了D-Gal诱导的肝衰竭模型的发生发展,有可能为急性肝衰竭的治疗提供一个新的治疗靶点。
Objective To study the devolpment of cardiotrophin-1 (CT-1 ) in acute liver failure rats (ALF) and to explore the clinical significance of it. Methods The model of ALF rats was induced by intraperitoneal injection with D-galac- tosamine (D-Gal), then their hepatic tissue samples were collected respectively on 6 h, 12 h ,24 h ,48 h ,72 h, 120 h, 168 h after D-Gal injection. The expression of CT-1 mRNA in hepatic tissue was detected by RT-PCR, and the expression of cox-2 protein was detected by immunohistoehemistry. Results CT-lmRNA in normal control group was hardly detected In ALF model group. But the expression of CT-lmRNA increased significantly at 12 hours after injection D-Gal(P 〈 0.05), and got peak value at 48 hours, then gradually decreased. By immunohistochemistry detection, COX-2 protein in normal group was negatively detected, Its expression increased gradually at 6 hours after injection D-Gal, reached the peak at 48- 72 h in ALF model group. Conclusion CT-1 was participating in the progress of the acute liver failure by D-galac- tosamine. It may become a new treat target in clinic.
