重组人乳铁蛋白在体外模拟胃肠环境中稳定性研究 被引量:4

摘要 目的研究重组人乳铁蛋白(recombinant human lactoferrin,rHLF)分别在体外模拟胃液和模拟肠液中的消化稳定性。方法采用模拟胃液和模拟肠液在体外建立模拟胃肠环境消化体系,测定重组人乳铁蛋白在胃肠环境中的稳定性。重组人乳铁蛋白在模拟胃、肠液中的浓度分别为5.0和2.0 mg/ml。在重组人乳铁蛋白与模拟胃/肠液反应后的0、15、30 s和1、2、5、10、20、30、60 min取样,根据SDS-PAGE凝胶电泳结果,判断重组人乳铁蛋白在模拟胃、肠液环境中的稳定性。结果重组人乳铁蛋白在体外模拟胃液中2 min内降解,在模拟肠液中60 min内不能完全降解。结论重组人乳铁蛋白在体外模拟人体胃肠环境中易被降解消化。 Objective To study the digestibility of recombinant human lactoferrin (rHLF) in simulated gastric fluid (SGF) and simulated intestinal fluid(SIF). Methods In in vitro simulated gastrointestinal environment of digestive system, test model of digestibility in SGF and SIF was established. The stability of recombinant human lactoferrin protein was tested. The concentrations of protein in SGF and SIF were 5.0 and 2.0 mg/ml respectively. At intervals of O, 15, 30 s and 1, 2, 5, 10, 20, 30, 60 rain, the samples were taken out. According to the results of SDS-PAGE, the stability of the protein to digestion in SGF/SIF was observed. Results The recombinant human lactoferrin protein was digested within 2 min in SGF and was not digested completely within 60 rain in SIF. Conclusion Recombinant human lactoferrin protein is considered instable in SGF/SIF.
