摘要
目的探讨伴生物质对山茱萸总苷提取物中马钱苷生物利用度的影响。方法以随机分组组间对照实验设计方法,采用HPLC法测定不同伴生物山茱萸总苷对大鼠灌胃给药40mg.kg-1后,不同时间点马钱苷的血药质量浓度,并以DAS2.0药动学软件计算相关参数。结果建立了马钱苷在血清中的HPLC分析方法,其灵敏度高,分离度和重复性好,可以满足本研究被测样品分析要求;不同伴生物山茱萸总苷提取物中马钱苷在大鼠体内的药物代谢动力学过程均符合二室模型,且随着样品中山茱萸总苷含量的升高,体内的消除速率和达峰质量浓度变化较大,山茱萸总苷Ⅳ在150min时达到峰质量浓度,而AUC(0-∞)有先减小后升高的趋势。结论山茱萸总苷中的伴生物质可能影响其中马钱苷体内的生物利用度。
Objective To study the associated substances of cornel total glycoside extract on loganin bioavailability of influence.Methods Randomized groups experiment design method was used,in which HPLC method for the determination of different associated substances was employed after administration of 40 mg·kg-1.DAS2.0 pharmacokinetic software was used to calculate relevant parameters.Result The established HPLC analysis method was sensitive,the separation degree and reproducibility was satisfactory,and the method can meet the requirements of sample analysis;Different associated substances of cornel total glycoside extract of loganin in rats in vivo were in line with the two compartment model.In vivo elimination rate and peak concentrations varied greatly.The cornel total glycoside Ⅳ in 150 min reached a peak concentration,and AUC(0-∞) were first decreased and then increased.Conclusion Associated substances of cornel total glycoside may affect the loganin in vivo bioavailability.
