摘要
目的对苯乙醇苷衍生物的辐射防护活性进行筛选。方法采用不同剂量X射线照射人皮肤成纤维细胞(Human Skin Fibroblasts,HSF),构建辐射损伤模型,以MTT法检测6种苯乙醇苷单体对HSF增殖的影响。结果与2 Gy、4 Gy和8 Gy辐射组比较,16 Gy辐射组对HSF增殖抑制作用的差异有统计学意义(P<0.01)。采用16 Gy一次性X射线照射构建辐射损伤模型,模型组与不同药物干预组在各时段比较差异均有统计学意义(P<0.01),其中毛蕊花糖苷辐射防护作用优于其他5种苯乙醇苷衍生物。X射线对HSF的增殖抑制作用与辐射剂量呈正相关。结论 X射线辐射对HSF具有明显损伤作用,而6种苯乙醇苷衍生物对其均具有一定的保护作用,其中以毛蕊花糖苷的抗辐射活性最强。
Objective To investigate the anti-radiation activity of phenylethanoid glycosides. Methods Human skin fibroblasts (HSF) were exposed to different dosages of X-ray to induce radiation injury model, and then MTT assay was used to detect the proliferation activity of HSF after treated by six kinds of phenylethanoid glycosides. Results The proliferation-inhibition effect of X-ray on HSF was positively correlated with the irradiated dose. The effect in 16 Gy group was more significant as compared with 2 Gy, 4 Gy and 8 Gy groups (P 〈 0.01 ), so one dose of 16 Gy X-ray was used to induce the radiation injury model. Compared with the model group, the six kinds of phenylethanoid glycosides showed obvious radiation prevention at all time points (P 〈 0.01 ), and acteoside showed better effect than the other five kinds of phenylethanoid glycosides. Conclusion X-ray irradiation can induce obvious damage to HSF. Phenylethanoid glycosides show certain anti-radiaton effect on HSF, and acteoside has the most potent anti-radiation effect in six kinds of phenylethanoid glycosides.
