摘要
为了开发一种廉价的培养基质以替代组培产业中相对昂贵的琼脂,本文利用一种特殊的稻米淀粉,研究了它在陆地棉胚珠离体培养中替代琼脂的效果。研究表明:在MS或B5培养基中只使用淀粉时,阻碍了纤维素的合成,引起胚珠褐化,但促进细胞分裂,尤其是在添加GA3时作用更明显;淀粉与琼脂混合不能促进纤维的伸长,但随淀粉比例的降低,胚珠纤维发生率增加,但只有在部分胚珠上能看到较短的纤维。
To develop a low price substrate to replace agar in tissue culture,we use a specially starch extracted from rice as a substrate and the effect of it's substitute for agar on upland cotton ovule culture were studied.The results showed,when once starch was solely used in either MS or B5 medium,it triggered the hindrance of cellulose biosynthesis and induced the phenolic browning on ovule though it had promoted cell division better than agar,particularly with GA3.As starch and agar combination did not well stimulate the growth of cultured fiber elongation until the subsequent decrease amount of starch substrate led to greater fiber responses;however,the fibers induced by starch and agar,visually expected,were frequently short and grew only some parts of ovules.
