摘要
【目的】观察高甘油三脂血症对急性坏死性胰腺炎(ANP)大鼠肿瘤坏死因子a(TNpa)、白介素-6(IL-6)表达的影响,探讨高脂血症相关性ANP的特性。【方法】SD大鼠54只,随机分成高脂血症+ANP模型组(A组),非高脂血症+ANP模型组(B组),高脂血症+假手术模型组(C组)。A、C组通过高脂饲料喂养3周成高甘油三脂血症模型,A、B组大鼠建立ANP模型。A、B组分别成模后4h、8h、12h时点测定血甘油三脂(TG)、血淀粉酶(AMS)、TNF-a和IL-6浓度,进行胰腺组织病理学评分;C组大鼠假手术建模后同样方法检测以上指标。【结果】①A组大鼠ANP成模前后TG浓度变化差异无统计学意义(P〉O.05),B组大鼠ANP成模后TG浓度较成模前增高(P〈0.05)。②A、B组大鼠ANP成模后TNF-a和IL-6随时点逐渐增高,且A组大鼠TNF-a和IL-6浓度明显高于B组(P〈0.05)。③A组大鼠4h、8h、12h时点胰腺组织病理学评分明显高于B组(P〈0.05),且与成模前TG浓度呈正相关。【结论】①高甘油三脂血症时TG的浓度与ANP胰腺病理损害存在相关性;ANP时可出现脂质代谢障碍。②高甘油三脂血症可上调ANP大鼠TNF-a和IL-6等炎症介质的释放。
[Objective] To observe the effect of hypertriglyceridemia on serum tumor necrosis factor-alpha(TNF-a) and interleukin-6(IL-6) in rats with acute necrotizing pancreatitis(ANP), and to explore the characteristics of hypertrig-lyceridemic-related ANP. [Methods] A total of 54 SD rats were randomly divided into hypertriglyceridemia+ ANP group (group A), ANP without hypertriglyceridemia group(group B) and hypertriglyceridemia+sham operation group(group C). Group A and group C were made to be high triglyeride(TG) model by feeding with high-fat diet for 3 weeks. Group A and group B were made to be ANP model. The levels of TG, AMY, TNF-a and IL-6 in group A and group B were measured at 4h, 8h and 12h after modeling. The pathology of pancreas was scored. Above indexes in group C after mod- eling of sham operation were measured by using the same method. [Results] In group A, there was no significant differ- ence in TG level between before and after the modeling( P 〉0. 05). Compared with before the modeling, TG level in group B after modeling increased( P 〈0.05). After modeling, the levels of TNF-a and IL-6 in group A and group B in- creased with the time gradually. The levels of TNF-a and IL-6 in group A were obviously higher than those in group B( P 〈0.05). The histological score of pancreas in group A at 4h, 8h and 12h was obviously higher than that in group B( P 〈0.05), and positively related with TG level before modeling. [Conclusion] The level of TG in hypertriglyceridemia is correlated with histological damage of pancreas of ANP. Lipid metabolism occurs in ANP. Hypertriglyceridemia can up regulate the release of inflammatory mediator such as TNF-a and IL-6 in ANP rats.
