摘要
为了检测哈萨克绵羊MHC抗包虫病基因型的抗性,了解肝包虫病在中间宿主绵羊体内发展过程中血液生理指标的变化规律,本实验筛选出4只健康的MHC抗包虫病基因型哈萨克绵羊作为抗性组,选取4只健康与包虫病抗性或易感性无关基因型的哈萨克绵羊作为非抗性组,模拟绵羊自然感染包虫病过程,采用口服细粒棘球绦虫虫卵的方法,建立绵羊自然感染包虫病的动物模型。同时采集攻虫后第9至16周、18周、20周、22周的绵羊血液,利用全自动血细胞分析仪测定血液生理指标。结果显示:在攻虫后白细胞和淋巴细胞变化趋势相似,总体水平均升高,且抗性组总体高于非抗性组,并在攻虫后第10周和第14周达到显著差异(P<0.05)。攻虫后,抗性组和非抗性组红细胞总数均逐渐升高,且抗性组的红细胞总数高于非抗性组,并在12-16周显著高于非抗性组(P<0.05)。整个试验过程中,抗性组粒细胞总数总体上高于非抗性组,并在攻虫后14-15周,显著高于非抗性组(P<0.05)。攻虫后红细胞分布宽度-SD和红细胞分布宽度-CV的变化趋势较为相似,且抗性组和非抗性组的差异较大,在攻虫后13、14、18周差异达到极显著水平(P<0.01),15、16周差异显著(P<0.05)。结果说明抗性组绵羊在攻虫后白细胞、淋巴细胞、红细胞、粒细胞等免疫相关细胞的先天免疫水平较高,表现出对包虫病有一定的抗性。对绵羊感染包虫病的血液生理指标变化趋势的跟踪监测为临床上诊断治疗该病提供了一定依据。
In order to investigate the resistance of MHC hydatidosis-resistant of Xinjiang Kazakh sheep and to understand the variation of blood physiological indices in the process of liver echinococcosis intermediate in host sheep,we chose four hydatidosis-resistant genotype sheep as the treatment group and another four non-resistant sheep as the control,simulated natural infection process of echinococcosis in sheep,fed them with hydatidosis eggs,and established animal model of natural echinococcosis-infection.After the infection,the blood was collected in wk 9-16,wk18,20 and 22 to analyze the blood indices with automatic blood cells analyzer.The result showed that:after the infection,the total white cell and lymphocyte in test group were higher than the control group,and the difference became significant in wk14-15(P0.05).In wk12-16,red blood cell count in test group was higher than the control;both of them were higher than before the infection.In the whole test process,total granulocyte in test group was significant higher than that in control(P0.05) in wk14-15.The variation of RDW-SD was similar to RDW-CV,and both of them in test group were significantly higher than in control(P0.01) in wk13,14 and 18,and significantly higher than in control(P0.05) in wk15 and 16.Conclusion:after infection,the resistant group has strong innate immune level in leukocyte,lymphocytes and red blood cells,showing a certain degree of echinococcosis-resistance.The variation of the blood physiological indices will provide the basis for the clinical diagnosis of echinococcosis.
