摘要
探索激活的细胞毒 T淋巴细胞 (CTL )的穿孔素途径 ,大鼠肝移植急性排斥的早期表达及诊断价值。在建立大鼠肝移植急性排斥模型的基础上 ,应用免疫组化和逆转录聚合酶链反应 (RT- PCR)检测移植肝穿孔素的表达 ,并以同基因肝移植为对照。急性排斥移植肝存在穿孔素的蛋白表达 ,术后 3天即急性排斥早期均表达穿孔素 4天后表达明显增强 ,并贯穿整个排斥反应全过程 ;而同基因对照组术后 7天内均不表达穿孔素。CTL 的穿孔素途径参与介导了大鼠肝移植急性排斥反应 。
To explore the early expression of perforin which is functional marker gene of activated cytotoxic T lymphocytes (CTL) in acute rejection of liver transplantation, the expression of perforin was examined by reverse transcription ploymerase chain reaction (RT PCR) and immunohistochemistry in hamster to rat liver grafts. Histological findings were studied comparatively and isografts as the control. The results showed that there were more perforin positive cells in the rejected grafts and perforin gene mRNA expressed at posttransplantation day (PTD) 3 in liver grafts, and the level of expression obviously increased after PTD 4, penetrating then through the whole course of acute rejection. The time of expression of perforin mRNA was at least 2 days earlier than that of pathological damage. Conclusions:The results strongly indicated that perforin played a role in cell mediated cytoysis of acute rejection in liver transplantation. Therefore, the expression of perforin mRAN may be valuable for early predictive diagnosis of acute rejection.
