安徽省新型布尼亚病毒基因测序鉴定方法的建立 被引量:1

Establishment of gene sequencing method for detecting severe fever with thrombocytopenia syndrome bunyavirus in Anhui province

摘要 目的建立发热伴血小板减少综合征布尼亚病毒基因测序鉴定方法,对安徽省在2011年从病人血清中分离到的10株疑似发热伴血小板减少综合征布尼亚病毒进行基因测序鉴定,并分析其序列特征。方法设计引物,RT-PCR法扩增病毒核酸,对扩增产物进行测序分析。结果设计的三对引物可有效扩增核酸序列,10株疑似发热伴血小板减少综合征布尼亚病毒L、M、S三段基因序列和2010年我国发现毒株序列相似性93%以上,10株病毒和Phlebovirus AH12、Phlebovirus HN6两株病毒同属于一个进化分支,和白蛉病毒属的其它病毒株不在一个分枝上。结论 2011年从病人血清中分离到的10株病毒为发热伴血小板减少综合征布尼亚病毒,应加强安徽省对该病的监测,有效预防大范围传播。 Objective To establish the gene sequencing method for detecting severe fever with thrombocytopenia syndrome bunyavirus(SFTSV),and to appraise the ten suspected virus strains from patient serum by analysis of its sequences in Anhui province in 2011.Methods Primers were designed,and the viral nucleic acid was amplified by RT-PCR method,and the product of the amplification DNA was sequenced.Results The three pairs of primers would be efficient amplification of nucleic acid sequences.The ten suspected virus strains were similar to SFTSV above 93%,and more closed related to Phlebovirus AH12 and Phlebovirus HN6,less related with other Phlebovirus.Conclusion The ten virus strains isolated from patient serum were severe fever with thrombocytopenia syndrome bunyavirus in 2011.So we should strengthen the disease monitoring in Anhui province,and make the effective prevention of itsspread.
