HPLC-ELSD分析测定贝母类药材中生物碱成分

Determination of Alkaloids in Fritillaria spp. by HPLC-ELSD Method

目的 :利用高效液相色谱法测定贝母类药材中生物碱成分。方法 :采用Silica 5 μm ( 4 6mm× 2 5 0mm)色谱柱 ,流动相 :A 环己烷 -乙酸乙酯 -二乙胺 ( 6∶4∶1) ,B 环己烷 -乙酸乙酯 -二乙胺 ( 6∶4∶0 12 ) ,流速 1 0mL·min-1,以ELSD - 5 0 0为检测器 ,外标法定量。结果 :贝母甲素在 0 75～ 3 0 0 μg范围内呈线性 ,回收率为97 2 1% (n =5 ) ,RSD为 1 5 8%。结论 :5种不同贝母药材的HPLC图谱有明显差异 ,本法为良好的测定贝母甲素的方法 。

Objective:To determine the content of alkaloid compound in Fritillaria by using a new HPLC-ELSD methods.Method:Alltech Silica column (5 μm,4 6 mm× 250 mm) was used, mobile phase A consisted of cyclohexane-acetic ester-ethylamine(6∶4∶1), mobile phase B consisted of cyclohexane-acetic ester-ethylamine(6∶4∶0 12), at 1 mL·min -1 flow rate. ELSD-500 detector was used for qualitative and quantitative analysis by external standard method.Results:Within 0 75～3 00 μg peimine had a good linearity. The average recovery was 97 21%( n =5) with RSD 1 58%.Conclusion:There are significant differences in HPLC trace among five species of Fritillaria , the method is sensitive and reproducible and it could be used for quantitative determination of Fritillaria drug.
