摘要
目的 :采用高效液相色谱法测定人血浆中酮洛芬浓度。方法 :用石油醚 -乙醚 ( 1∶1)提取血浆样品中酮洛芬及萘普生 (内标 ) ,在SpherisorbC18柱上 ,以甲醇 - 5 0mmol·L-1磷酸二氢钠 ( 63∶3 7)为流动相进行分离 ,于 2 5 5nm检测。结果 :线性范围为 0 0 4~ 12 μg·mL-1,萃取回收率为 85 2 %~ 89 9% ,加样回收率为 97 0 %~ 10 0 1% ,日内RSD为 0 4 5 %~ 2 72 % ,日间RSD为 1 3 1%~ 4 4 0 %。结论 :本法简便 ,准确 ,重现性好 ,用于测定
Objective:A simple HPLC method is established for the determination of ketoprofen in human plasma.Method:Ketoprofen and the internal standard(naproxen)were first extracted into the mixture of petroleum ether and ether and then separated on C 18 column with a mobile phase consisting of methanol and 50 mmol·L -1 phosphate buffer(63∶37).Result:The calibration curve was linear within 0 04~12 μg·mL -1 .The extraction recoveries of ketoprofen were 85 2%~89 9%.The recoveries of methodology were 97 0%~100 1%.Inter-day and intra-day RSD were 0 45%~2 72% and 1 31%~4 40% respectively.Conclusion:This method is simple,accurate and reproducible.It has been applied successfully to the determination of ketoprofen in human plasma for pharmacokinetic studies.
