RP-HPLC法测定贯叶连翘片剂中有效成分金丝桃素和金丝桃苷的含量

Determination of Effective Compounds Hypericin and Hyperoside in Hypericum perforatum Tablets by RP-HPLC

目的 :采用RP -HPLC法测定贯叶连翘片剂中有效成分金丝桃素 (hypericin)和金丝桃苷 (hyperoside)的含量。方法 :分别使用ODS和ODS - 2色谱柱 ,流动相分别是 1 5 6%磷酸二氢钠 (用磷酸调 pH至 2 1) -甲醇 -醋酸乙酯 ( 1∶4∶1)及甲醇 - 0 5 %磷酸 ( 4 5∶5 5 ,用三乙胺调 pH至 3 0 ) ,检测波长分别为 5 90nm ,3 60nm。结果 :加样回收率分别为 98 3 2 % (n =5 ) ,96 0 4 % (n =5 )。金丝桃素的进样量为 0 0 0 98～ 0 16μg、金丝桃苷的进样量为 0 4 3～ 3 4 2 μg时 ,线性关系良好。结论

Objective:To determine hypericin and hyperoside, which are two effective constituents of Hypericum perforatum by RP-HPLC.Method:Hypericin and hyperoside were separated by ODS column with 1 56 % sodium dihydrogen phosphate (adjusted to pH=2 1 with phosphoric acid)-methanol-ethyl acetate(1∶4∶1) as mobile phase and ODS-2 column with methanol-0 5% phosphoric acid(45∶55,adjusted to pH=3 0 with triethylamine) as mobile phase, and detected with UV/ vis at 590 nm and 360 nm respectively.Result:The linear ranges of hypericin and hyperoside were 0 009 8～0 16 μg and 0 43～3 42 μg, respectively. The recoveries of them were 98 32% ( n =5) and 96 04% (n =5), respectively.Conclusion:These methods had good reproducibility and sensitivity.
