摘要
单链抗体 (Sc Fv)是用基因工程方法将抗体重链可变区 (VH)和轻链可变区 (VL)通过一段连接肽 (L inker)连接而成的重组蛋白 ,是保持了亲本抗体的抗原亲和活性和特异性的最小功能性抗体片段 .与完整抗体 Ig G及抗体 Fab段相比 ,具有在非靶向组织中滞留时间短、免疫原性低、血液清除快、肿瘤穿透力强、可在细菌中很经济地生产并可用基因工程的方法构建与其他效应分子连接的抗肿瘤融合蛋白等优点 .因此 Sc Fv成为将药物、毒素、放射性毒素导向肿瘤的很有价值的分子 。
Functional single chain antibodies (scFvs) are engineered by linking together immunoglobulin heavy and light chain variable domains (V H and V L) via polypeptide tethers and are the smallest functional fragments of antibodies that maintain the binding and specificity of the whole antibody. Compared with intact IgG or Fab fragments, scFvs have shorter retention time in non target tissues, lower immunogenicity, more rapid blood clearance and better tumor penetration. They can be produced economically in bacteria and can be manipulated by genetic engineering to form anti tumor fusion proteins incorporating additional effects or functions. ScFvs therefore represent valuable molecules for the targeted delivery of drugs, toxins or radionuclides to a tumor site and have great potential for the development of therapeutic or diagnostic reagents.
