摘要
目的:检测冠状动脉微栓塞后Toll样受体4(Toll-like receptor 4,TLR4)的水平。方法:以前降支内注射微球的方法建立的猪冠状动脉微栓塞模型为实验组(n=6);以前降支内注入0.9%氯化钠液的猪为对照组(n=6)。用蛋白质印迹(West-ern-blot)和实时聚合酶链反应(real-time polymerase chain reaction)方法检测对照组和实验组心肌标本前、后壁的TLR4水平。结果:实验组前、后壁TLR4蛋白水平分别为(0.46±0.02)、(0.17±0.01),而对照组前、后壁分别为(0.17±0.01)、(0.16±0.01)。实验组前、后壁TLR4 mRNA水平分别为(3.14±0.05)、(1.17±0.02),而对照组前、后壁分别为(1.25±0.01)、(1.03±0.03)。实验组后壁TLR4蛋白和mRNA水平与对照组相比无显著差异,而实验组前壁显著高于实验组后壁及对照组前后壁。结论:冠状动脉微栓塞后受累心肌组织的TLR4表达增加。
Objective:To study the expression of Toll like receptor 4 (TLR4) in swine coronary microembolization (CME) models. Methods:CME models were made by injection of inertia plastic microspheres (diameter 42μm,) into left anterior de seending coronary in swines. TLR4 in myocardial tissues of anterior wall and posterior wall was detected using Western-blot and real-time polymerase chain reaction. Results: The TLR4 protein levels of anterior wall and posterior wall were (0.46 ± 0.02) and (0.17±0.01) in study group, and those were (0.17±0.01) and (0.16±0.01) in control group. TheTLR4 mRNA levels of anterior wall and posterior wall were (3.14± 0.05) and (1.17± 0.02) in study group, and those were (1.25± 0.01) and (1.03 ±0.03) in control group. The expression of TLR4 was significantly higher in the anterior wall than the posterior wall in study group; and there were no significant difference in the expression level of TLR4 among the anterior wall, posterior wall in control group and the posterior wall in study group. Conclusions: The expression of TLR4 is elevated in the affected myocardial tissue after CME.
