碳铂壳聚糖微球瘤内化疗的实验研究 被引量:6

Intratumoral chemotherapy via chitoson polymer microsphere carried with carboplatin to treat C 6 glioma in rats

摘要 目的 建立大鼠的C6胶质细胞瘤模型 ,对碳铂壳聚糖缓释微球瘤内化疗的疗效进行研究。方法 通过大鼠的C6胶质细胞瘤模型 ,用碳铂壳聚糖缓释微球进行瘤内化疗 ,检测药物的疗效和动力学分布。结果 随着药物从微球中的释放 ,药物的浓度逐渐上升 ;瘤心浓度最高 ,边缘次之 ,瘤旁脑组织更低 ;碳铂在肾脏及血浆内的分布是一致的 ,但较其在脑内的浓度相差甚远 (P <0 0 0 0 1) ;病理检查显示投药后肿瘤细胞的生长受到明显的抑制 ,并有明显的剂量依赖性 ,Giemsa染色肿瘤细胞出现特征性的凋亡细胞群。结论 碳铂壳聚糖缓释微球有效地克服了纯碳铂制剂的不足 ,瘤内化疗效果佳 ,全身、局部毒副反应均小 ,具有很好的临床前景。 Objective To use chitoson polymer microsphere carried with carboplatin to treat C 6 glioma in rats, and demonstrate the effect of controlled release drug for intratumoral chemotherapy.Methods The C 6 glioma model in rats was established for intratumoral chemotherapy with chitoson polymer microsphere carried with carboplatin. The pharmacodynamics and pharmacokinetics of chitoson biodegradable polymer microsphere carried with carboplatin were investigated.Results The concentration of carboplatin increased gradually after interstitial delivery from the biodegradable polymer implant in the rats with C 6 glioma. The concentration of carboplatin in center of tumor is the highest that of, the border of tumor is lower, ant that of the tissue beside tumor is the lowest (P<0 001). The distribution of carboplatin in renal and blood plasma is consistency, but much lower than brain tissue (P<0 0001). Pathological examination reveals that tumor growth was inhibited significantly after drug implant without any obvious neurotoxicity. G staining demonstrated the existence of tumor apoptosis.Conclusions The chitoson polymer microsphere carried with carboplatin has an excellent result in intratumoral treatment of C 6 glioma in rats. It provides us a promising approach to treat intracranial malignant tumor.
