摘要
目的研究血必净对博莱霉素诱导的肺纤维化大鼠IL-1β、IL-6、IL-13及TNF—α表达及肺组织病理改变的影响。方法将64只Wistar大鼠随机分成空白组、模型组、血必净组及甲泼尼龙组,每组16只。除空白组外,其余3组以气管内滴注博来霉素(5mg/kg)建立纤维化模型,于造模后第14、28天取材。采用HE及Masson染色法观察肺组织病理变化,酶联免疫吸附(ELISA)法检测血清IL-1β、IL-6、IL-13及TNF-α的含量。结果在第14天时,血必净组肺泡炎程度明显轻于模型组,大鼠肺泡炎积分明显降低(P〈0.05),血清IL-13较模型组明显降低(P〈0.05),TNF—α较模型组明显降低(P〈0.05);在第28天时,血必净组仅形成轻中度肺间质纤维化,肺纤维化积分较模型组明显降低(P〈0.05)。结论血必净能显著降低血清IL-13及TNF-α表达水平,减轻肺组织肺泡炎及肺纤维化程度,从而抑制或延缓博莱霉素致大鼠肺纤维化的发生、发展。
Objective To investigate the effect of Xuebijing on serum cytokine contents and pathological changes in the lung of rats with bleomycin-induced pulmonary fibrosis. Methods Sixty four Wister rats were randomly divided into four groups (n=16 in each group): control group, model group, the xuebijing group and methylprednisolone group. The pulmonary fibrosis model was induced by intra-tracheal injection of bleomycin. The animals were sacrificed d14 and d28, the lung tissue was examined by HE staining and Masson staining and the serum contents of IL-1β、IL-6、IL-13、TNF-α were assayed by ELISA. Results Compared with model group, the serum levels of IL-β and TNF- α, and the degrees of alveolitis decreased significantly in Xuebijing group at d14 (P〈0.05); the pulmonary fibrosis was attenuated at d28 (P〈0.05). Conclusion Xuebijing can attenuate the alveolitis and pulmonary fibrosis, and decrease serum contents of IL-13 and TNF- α in bleomycin-induced pulmonary fibrosis rats.
