胃癌患者间隙性连接蛋白37基因C1019T多态性与幽门螺杆菌感染的相关性 被引量:3

Association between C1019T polymorphism of Connexin 37 gene and Helicobacter pylori infection in patients with gastric cancer

摘要 目的研究胃癌患者间隙性连接蛋白(Connexin)37基因C1019T多态性与幽门螺杆菌(Hp)感染的相关性。方法选取胃癌患者388例(胃癌组)及慢性浅表性胃炎患者204例(对照组)。胃黏膜活检组织切片染色法检测Hp感染情况,基因测序技术检测Connexin37基因C1019T多态性。比较基因型及等位基因分布频率。结果(1)Connexin37基因共有CC型、TC型、TT型3种基因型,在两组中的分布均符合Hardy—Weinberg遗传平衡规律。(2)胃癌组C等位基因分布频率显著高于对照组(P〈0.01)。与TT纯合子相比,CC+TC基因型胃癌患病风险显著增加(OR=2.47、95%CI=1.68~361,P〈0.01)。(3)分层分析提示Hp阳性是男性患者发生胃癌的一个危险因素,且男性Hp阳性者发生胃癌风险明显高于女性。剔除性别影响后,Hp阳性与胃癌密切相关(OR=8.82、95%CI=5.45~14.28,P〈0.01)。(4)胃癌组中Hp感染者C等位基因分布频率显著高于非感染者(P〈0.01)。与TT纯合子相比,CC+TC基因型胃癌患病风险显著增加(OR=2.96、95%CI=1.76~2.99,P〈0.01)。结论胃癌患者Connexin37基因T等位基因与胃癌的发生有关,且与Hp感染相关。 Objective To investigate the association between Connexin 37 C1019T polymorphism and Helicobacter pylori (Hp) infection in patients with gastric cancer. Methods Three hundred and eighty eight patients with gastric cancer (GC) and 204 patients with chronic superficial gastritis (CSG) were enrolled in the study. H. pylori was detected by gastric mucosal biopsies biopsy dyeing method. Genotype of Connexin37 gene 1019 sites was detected by gene sequencing technology, and genotypes and alleles frequencies were compared between GC and CSG groups. Results There were 3 genetic types (CC, TC, TT) in Connexin37 gene 1019 sites.The distribution frequency in two groups were consistent with the Hardy-Weinberg ge- netic balance rule. In comparison with CSG group, the frequency of Connexin37 C allele was higher in GC group(P〈0.01 ).The risk of gastric cancer prevalence was significantly increased in the carriers with C allele (CC+TC) than that with TT homozygote (OR=2.47, 95%C1=1.68- 3.61,P〈0.01). Hierarchical analysis showed that HP positive males had higher risk of gastric cancer than females. Elimination of gender effects, positive Hp and gastric cancer were closely correlated (OR=8.82, 95% CI =5.45-14.28, P〈0.01). The distribution frequency of C allele in patients with Hp infection was much higher than that in Hp negative ones in GC group (P〈0.01). Compared with TT homozygotes the risk of gastric cancer in CC+TC genotype increased significantly (OR=2.96, 5%Cl=1.76N2.99, P〈0.01). Conclusion The T allele in the Connexin37 gene might be associated with gastric cancer and with Hp infection.
