rhTFAR19蛋白质对γ射线诱导MCF-7细胞周期及凋亡影响的初步研究 被引量:35

摘要 目的 探讨细胞核内蛋白TFAR19对γ射线诱导的MCF 7细胞周期及凋亡的影响。方法 用细胞形态学、荧光显微镜及激光共聚焦荧光显微镜、单细胞微凝胶电泳、流式细胞仪进行观察和检测。结果 rhTFAR19蛋白对γ射线诱导MCF 7细胞周期无明显作用。细胞周期特异性凋亡为G2 M期细胞凋亡。凋亡细胞形态表现为核碎裂 ,染色质边集 ,凋亡小体形成。单细胞电泳可显示典型的“彗星” ,并通过形态变化区别坏死细胞和辨别荧光显微镜下难以识别的核碎裂凋亡细胞。结论rhTFAR19蛋白对γ射线诱导的MCF 7细胞周期无明显影响 ,其对细胞凋亡的促进作用可能是通过放大或增强γ射线诱导细胞凋亡的信号产生。 Objective To investigate the effect of protein product of TFAR19, an apoptosis related gene previously cloned from leukemia cell line TF 1 cultured in medium deprived of growth factor, on cell cycle and apoptosis of human breast cancer induced by γ ray.Methods Cell morphology, fluorescence microscopy, laser scanning confocal microscopy, single cell gel electrophoresis (SCGE) and flow cytometry (FCM) were applied to study cell cycle and apoptosis of breast cancer cell line MCF 7.Results rhTFAR19 protein itself had no effect on cell cycle arrest and apoptosis of MCF 7 cell induced by γ ray. When MCF 7 cells were pretreated with 10 μg/ml rhTFAR19 and then irradiated with 12 Gy γ ray, cell apoptosis was observed. It was characterized morphologically by chromatin condensation, nucleoar disintegration, formation of apoptotic bodies, and typical “comet” on SCGE.Conclusion TFAR19 protein has no apparent effect on MCF 7 cell cycle induced by γ ray. However, it synergizes with γ radiation in the induction and enhancement of apoptosis. Subject
