摘要
目的考察丹参及其化学成分对人粘连组织成纤维细胞(ATF)MMP-1与TIMP-1 mRNA表达的影响。方法使用组织块培养的方法获取ATF,分别加入丹参提取物、丹参素钠、丹参酮ⅡA磺酸钠与地塞米松培养48 h,提取总RNA并逆转录为cDNA,采用Real-time PCR法检测各组MMP-1与TIMP-1 mRNA的表达水平。结果与空白对照组比较,丹参提取物与丹参素钠10、40μg.mL-1剂量组MMP-1 mRNA水平差异有统计学意义(P<0.01),丹参素钠40μg.mL-1剂量组TIMP-1 mRNA水平差异有统计学意义(P<0.01),其余各组差异均无统计学意义(P>0.05)。结论上调MMP-1 mRNA水平是丹参抑制ATF胶原过度合成,防止粘连产生的作用机制之一,丹参素在其中起主要作用。
Objective To study the effect of Salvia miltiorrhiza and its chemical compositions on the expression of MMP-1 and TIMP-1 mRNA in human adhesion tissue fibroblasts(ATF).Method ATF from human peritoneal adhesions were harvested by primary culture.The expression of MMP-1 and TIMP-1 mRNAs in ATF treated with Salvia miltiorrhiza extract,sodium danshensu,and sodium tanshinone ⅡA sulfonate were determined by real-time PCR.Dexamethasone was used as positive control.Results Compared with the control group,the levels of MMP-1 mRNA increased significantly in the animal treated with Salvia miltiorrhiza extract and sodium danshensu at the dosage of 10 or 40 μg·mL-1(P0.01).Meantime,the levels of TIMP-1 mRNA enhanced markedly in the animal treated with sodium danshensu at the dosage of 40 μg·mL-1(P0.01).There were no difference between the control and other groups(P0.05).Conclusion Salvia miltiorrhiza could upregulate MMP-1 mRNA expression in ATF,which inhibit the over synthesis of collagen and prevent the formation of adhesion.Danshensu play a major role in this process.
