摘要
目的:建立贯叶金丝桃中芦丁与金丝桃苷的含量测定方法。方法:采用高效液相色谱仪,使用Diamonsil C18柱(250mm×4.6 mm,5μm),流动相为甲醇-乙腈-四氢呋喃-0.4%醋酸(1∶1∶18∶80),流速为0.7 ml·min-1,检测波长为358 nm,柱温为30℃。结果:芦丁在0.206~1.236μg范围内有良好的线性关系,r=0.999 9。金丝桃苷在0.144~0.864μg范围内有良好的线性关系,r=0.999 8;芦丁的平均加样回收率为98.57%,RSD为0.94%,金丝桃苷的平均加样回收率为98.57%,RSD为0.94%。结论:该方法简便快捷,准确度高。
Objective: To establish an HPLC method for the determination of rutin and hyperoside in Hypericum perforatum L..Method: A Diamonsil C18 column(250 mm×4.6 mm,5 μm) was eluted by the mobile phase consisted of methanol-acetonitrile-tetrahydrofuran-0.4% acetic acid(1∶ 1∶ 18∶ 80) at the column temperature of 30℃.The flow rate was 0.7 ml·min-1 and detection wavelength was 358 nm.Result: Rutin was linear within the range of 0.206-1.236 μg with a regression coefficient of 0.999 9,and the average recovery was 98.79% with RSD of 0.91%.Hyperoside was linear within the range of 0.144-0.864 μg with a regression coefficient of 0.999 8,and the average recovery was 98.57% with RSD of 0.94%.Conclusion: The method is quick,simple and sensitive.
