摘要
目的:建立尿素维E软膏中主药含量测定方法。方法:采用HPLC法,尿素的色谱条件:色谱柱:依利特HypersilNH2柱(250 mm×4.6 mm,5μm),流动相:乙腈-水(95∶5),流速:1.0 ml·min-1,检测波长:190 nm,柱温:25℃;维生素E的色谱条件:色谱柱:依利特Hypersil ODS柱(250 mm×4.6 mm,5μm),流动相:甲醇-异丙醇(80∶20),流速:1.0 ml·min-1,检测波长:285 nm,柱温:25℃。结果:尿素含量在100~600μg·ml-1范围内线性关系良好(r=0.999 9),平均回收率为100.5%,RSD为1.4%(n=9)。维生素E含量在8~40μg·ml-1范围内线性关系良好(r=0.999 9),平均回收率为99.6%,RSD为1.0%(n=9)。结论:该方法操作快速、准确、专属性和重复性好,可用于尿素维E软膏中尿素和维生素E的含量测定。
Objective: To establish the content determination method for urea vitamin E ointments.Method: The urea was detected on a Hypersil NH2 column(250 mm×4.6 mm,5 μm) with acetonitrile-water(95∶ 5) as the mobile phase,the flow rate was 1.0 ml·min-1 and the detection wavelength was 190 nm.Vitamin E was detected on a Hypersil ODS column(250 mm×4.6 mm,5 μm) with methanol-dimethyl carbinol(80∶ 20) as the mobile phase,the flow rate was 1.0 ml·min-1 and the detection wavelength was 285 nm.Result: The linear range was 100-600 μg·ml-1(r=0.999 9) for urea with the average recovery of 100.5% and RSD of 1.4%(n=9).The linear range of vitamin E was 8-40 μg·ml-1(r=0.999 9) with the average recovery of 99.6% and RSD of 1.0%(n=9).Conclusion: The method is simple,quick and accurate,and can be used in the content determination of urea vitamin E ointments.
