摘要
目的探讨NKG2D、MICA和IL-8在早期自然流产(ESA)患者蜕膜绒毛中的表达及其意义。方法选取20例ESA患者作为流产组,20例正常早孕人工流产妇女作为对照组,RT-PCR和Western Blotting法检测两组蜕膜绒毛组织中NKG2D、MICA和IL-8mRNA及蛋白的表达,免疫组织化学法检测蜕膜绒毛组织中MICA蛋白的表达,分析NKG2D、MICA与IL-8之间的相关性。结果流产组蜕膜绒毛组织中NKG2D、MICA和IL-8mRNA表达水平低于对照组;流产组蜕膜绒毛组织中NKG2D、MICA和IL-8蛋白表达低于对照组,差异均有统计学意义。MICA存在于绒毛组织合体滋养层细胞、细胞滋养层细胞及蜕膜组织基质细胞中。NKG2D、MICA与IL-8mRNA呈正相关(r=0.71、0.74,P〈0.05);NKG2D、MICA与IL-8蛋白呈正相关(r=0.76、0.77,P〈0.05)。结论ESA患者蜕膜绒毛组织中NKG2D、MICA和IL-8mRNA和蛋白表达水平低,这可能是导致滋养细胞侵袭性降低并发生ESA的重要原因。
Objective To investigate he expression and implication of natural-killer group 2 member D (NKG2D), MHC class I chaln-related molecule A (MICA), and Interleukin-8 (IL-8) in villi and decidua of first trimester spontaneous abortus. Methods Expression of NKG2D, MICA, and IL-8 mRNA and protein in villi and decidua of first trimester spontaneous abortus ( abortion group, n = 20) and normal first trimester pregnant women ( control group, n = 20) was detected by semi-quantitative RT-PCR and Western blot. MICA protein was detected by immunohistochemistry. Results Expression of NKG2D, MICA, and IL-8 mRNA was significantly lower in villi and decidua in the abortion group compared with that in the control group (P 〈 0.05 ). Expression of NKG2D, MICA, and IL-8 protein in villi and decidua in the abortion group was significantly lower than that in the control group ( P 〈 0.05 ). Expression of NKG2D and MICA in villi and decidua at first trimester has significantly positive correlation with the expression of IL-8. Immunohistochemistry showed that MICA existed in the villous syncytiotrophoblast cells and decidual stromal cells. Conclusion Lower expression of NKG2D and IL-8 in villi and decidua of early spontaneous abortus may be associated with the decrease in the ability of early embryo invasion. It may lead to spontaneous abortion during early pregnancy.
