实验性癫痫大鼠致痫后脑组织中MDA、SOD、CAT变化的研究

摘要 目的通过研究实验性癫痫大鼠在癫痫发作后不同时间脑组织中MDA、SOD、CAT的动态变化探讨其与癫痫的关系。方法选取健康雄性Wistar大鼠随机分成2组。A组(生理盐水组),B组(硫代氨基脲致痫组)。B组再随机分成5组,每组各5只。B1组:致痫后0h;B2组:致痫后2h;B3组:致痫后1d;B4组:致痫后3d;B5组:致痫后7d。分别采用硫代巴比妥酸法、羟胺法、紫外分光法测定各组脑组织中的MDA、SOD、CAT。结果致痫后0h组与生理盐水组比较,MDA极显著增高,SOD、CAT极显著降低(P〈0.001);致痫后2h组与生理盐水组比较,MDA非常显著升高,SOD、CAT非常显著降低(P〈0.01);致痫后1d组与与生理盐水组相比,MDA显著升高,SOD、CAT显著降低(P〈0.05);致痫后3d组和致痫后7d组与生理盐水组比较,无明显统计学差异(P〉0.05)。结论实验性癫痫大鼠在致痫后自由基参与了其病理发生、发展过程,MDA可能反映了脑损害程度,SOD、CAT可能反映了脑保护作用。 Objective To observe the changes of MDA,SOD and CAT in brain tissues of experimental epileptic rats in different times after epilepsy seizure.Methods Wistar rats were divided into two groups,control group(Group A),group injected with N.S and group injected with thiosemicarbazide(Group B).The test of MDA in brain tissue use thibarbituric acid method.The test of SOD use hydroxylamine method and the test of CAT use ultraviolet spectroscopy method.Results Group B 1 (0 hour after epilepsy seizure) were greatly significantly increased as compared with Group A,SOD and CAT of Group B1 were greatly reduced(P 〈 0.001).MDA of Group B2(2 hours after epilepsy seizure) were quite significantly increased as compared with Group A,SOD and CAT of Group B2 were quite reduced(P 〈 0.01).MDA of Group B3(1 day after epilepsy seizure) were significantly increased as compared with Group A,SOD and CAT of Group B3 were significantly reduced(P 〈 0.05).The MDA,SOD and CAT of Group B4(3 days after epilepsy seizure) and group B5(7 days after epilepsy seizure) weren't different as compared with Group A in statistics(P 〉 0.05).Conclusion Free radical could join the pathological passage of experimental epiliptic rats after seizure:MDA could suggest cerebral injury effect,SOD and CAT could indicate cerebral protective effect.
