摘要
目的:建立内消旋玉米黄素含量的手性HPLC测定方法。方法:色谱柱为CHIRALPAK手性柱(4.6×250mm,5μm),流速为0.5mL/min,检测波长453nm,柱温为30oC,进样量为20μL,流动相为正己烷(溶剂A)和丙酮(溶剂B),采用梯度洗脱,0-60min溶剂A由90%变为80%、溶剂B由10%变为20%,然后保持80%的溶剂A和20%的溶剂B保持10min。结果:内消旋玉米黄素在50~5252ng/mL浓度范围内与峰面积呈良好的相关性(r=0.9997),平均回收率为99.2%,RSD为0.4%,最低检出限为1 1.5ng/mL。结论:本方法专属性强、灵敏度高、重现性好、结果准确,适用于内消旋玉米黄素的质量控制。
Objective To establish a chiral - HPLC method to determine the content of meso - zeaxanthin.Methods The determination was performed on an CHIRALPAK column(4.6 x 250mm,5μm),the flow rate was 0.5mL/min and the wavelength of detection was at 453nm,the column temperature was 30℃,the injection volume was 20μL,mobile phase was hexane(solvent A) and acetone(solvent B).The elute was first by 90%solvent A then change to 80% solvent A,and 10%solvent B first and then change to 20%solvent B over a 60min linear gradient;and then kept with 80%solvent A and 20%solvent B for 10 min.Results:A good linearity between the concentrations of the samples and their peak areas was shown from 50 to 5252 ng/mL(r =0.9997).The average recovery rate was 99.2%,RSD was 0.4%(n =9),and the minimal detectable concentration of meso - zeaxanthin was 11.5ng/mL.Conclusions The method is accurate and sensitive.It can be used for quality control of meso - zeaxanthin.
