摘要
目的:探讨依达拉奉对脓毒症大鼠心肌组织的肿瘤坏死因子α(TNF-α)和一氧化氮(NO)的影响。方法:54只SD大鼠随机分成对照组、脓毒症组、治疗组,建立腹腔注射脂多糖大鼠脓毒症模型(LPS 10 mg/kg),治疗组予以腹腔注射LPS 10 mg/kg和静脉注射依达拉奉3 g/kg,每组按造模后6h、12h、24h随机分成三个亚组,检测不同时点各组大鼠血清肌钙蛋白T(CTnT),心肌组织肿瘤坏死因子α(TNF-α)、一氧化氮(NO)含量,以及24 h后心肌细胞显微镜下改变。结果:在6 h时、12 h时和24 h时点各组血清中CTnT浓度、心肌匀浆TNF-α和NO含量,脓毒症组>治疗组>对照组,差异有统计学意义(P<0.05)。脓毒症组和治疗组心肌匀浆中的TNF-α在6 h时达到高峰,随着建模时间的延长,心肌匀浆中的TNF-α逐渐回落;同一组内各时点之间比较,差异有统计学意义(P<0.05)。显微镜显示治疗组与脓毒症组相比,心肌细胞断裂、溶解减轻,间质水肿明显改善。结论:依达拉奉可通过减少心肌TNF-α、NO生成对脓毒症大鼠心肌组织发挥一定的保护作用。
Objective :To explore the effects of edaravone on TNF-α and NO in septic rat's myocardial tissue. Method:54 SD rats were randomly divided into the control group, the septic group and the treatment group. The septic group was intraperitoneal injected with Lipopolysaccharide (LPS)(10 mg/kg). The treatment group was intraperitoneal injected with edaravone (3 g/kg). The control group was injected with saline. The three groups were randomly divided into three subsets (6 rats each) and killed at 6h, 12h and 24h respectively. The change of CTnT in blood samples,TNF-α and NO of myocardial tissue were detected at different points. The pathological changes of heart were observed by light microscope. Results: The CTnT in serum was statistically different at 6 h,12 h and 24 h, sepsis group 〉 treatment group 〉 control group(P 〈 0.01). The TNF-α and NO of myocardial tissue in the septic group significantly increased, compared to the treatment group (P 〈 0.01 ) and control group (P 〈 0.01 ), and it significantly increased in the treatment, compared to control group (P 〈 0.05) at 6h, 12h and 24h after operation. The TNF-α of myocardial tissue in the septic group and the treatment group achieved the peak value at 6h and decreased gradually. There were significant differences between each other (P 〈 0.05). Pathological examination of heart by light microscope in the septic group, the cardiomyocyte arranged disorderly, fractured and lised. Myocardium became acidophilic, myocardial tissue was edematous and neutrophil infiltrated. The treatment group significantly improved in pathological examination of heart, compared to the septic group. Conclusion: Edaravone protected the septic rat's myocardial injury, one probable mechanism is reducing the expression of TNF-α, NO in heart.
